The internal positive control is supplied by a plasmid encompassing a sequence similar to the assay target sequence except for the probe hybridization site. The multiplex qPCR is performed with both probes and one set of outside amplification primers. The PCR products are monitored by using a single excitation source at 470 nm (laser of LED) and emissions at 530 nm from FAM, indicating control product, or 705 nm from Cy5.5 by FRET from FAM, indicating assay product.
