Standard DNA Synthesis CPG

High loading, convenient pore size: all regular synthesis CPG beads are conveniently offered at uniform 1000A that is suitable for oligos for as long as 120 nt as tested by Allele, while not sacrificing loading capacity—42-45 micromol/gram, a range normally achievable by using 500A CPG beads. Oligo Modification CPG

3’-Amino-Modifier C7 CPG

  • introduces a primary amine to functionalize the 3’-terminus of the target oligonucleotide
  • can be use to attach a variety of modifiers (i.e., activated NHS ester or isothiocyanate fluorescent label) to an oligo
  • can be used to attach an oligo to a solid surface
  • users may print their own oligonucleotide-based microarrays

3’-Biotin CPG:

  • can be used to detect proteins using various methods, i.e. non-radioactive immunoassays, cytochemical staining, cell separate, isolation of nucleic acids, detection of specific DNA/RNA sequences by hybridization, probing of conformational changes in ion channels
  • provides means for purification

3’-Dabcyl CPG

  • a commonly used dark quencher due of its light absorbance properties and lack of residual fluorescence
  • dabcyl’s absorption properties limit the range of fluorophores it can quench to those emitting at 400-550 nm with max absorption at 471 nm
  • doubly labeled oligonucleotides can be used as diagnostic probes for DNA and RNA quantification and to discriminate single nucleotide variations

3’-(6-FAM) CPG

  • label oligos with fluorescein, a fluorophore with an isosbestic point (equal absorption for all pH values) at 460 nm, at the 3’-end
  • fluorescein has an absorption maximum at 494 nm and emission maximum of 521 nm
  • doubly labeled oligonucleotides can be used as diagnostic probes for DNA and RNA quantification and to discriminate single nucleotide variations
  • singly labeled oligonucleotides can be used for DNA sequencing and in situ hybridization

3’-Thiol-Modifier C6 S-S CPG

  • important in the production of diagnostic probes
  • terminal thiol group allows attachment of an oligo to various ligands (i.e., biotin, alkaline phosphatase, or fluorescent dyes)
  • allows for the attachment of an oligo to a solid surface via a maleimide linkage

 

Oligo Modification Phosphoramidites

5'-FAM Phosphoramidit

  • can label the 5’-end of an oligo with a fluorescein, a fluorophore with an isosbestic point (equal absorption for all pH values) at 460 nm
  • fluorescein has an absorption maximum at 494 nm and emission maximum of 521 nm
  • doubly labeled oligonucleotides can be used as diagnostic probes for DNA and RNA quantification and to discriminate single nucleotide variations
  • singly labeled oligonucleotides can be used for DNA sequencing and in situ hybridization
