Immunohistochemical staining is widely used as a diagnostic tool to visualize abnormal cells such as those found in cancerous tumors. Specific antibody are used to binds to its target antigens in wax or cryo tissue sections. This technique enables the researcher to gain better understanding of a particular protein distribution and localization of biomarkers and differentially expressed proteins in different part of the biological tissue. This technique can be done using either traditional brightfield microscopy which utilized 3,3-Diaminobenzidine (DAB - brown) to visualize the immuno products and Mayers’ hematoxylin is commonly used as a nuclear counterstain or immunofluorescent microscopy where a fluorescent protein is used and 4',6-diamidino-2-phenylindole (DAPI) as a counterstain.
